you leave 90% of the atmosphere behind you and all the air breathing
hardware.

the actual manned space part would be a small winged vehicle

release would be slow. why go fast all the way?

manned mini shuttle would be rocket powered

There's not a lot (some, but not a lot) to be gained by releasing a
second stage launch vehicle from high altitude but at subsonic speed,
and it doesn't take that long to reach the maximum feasible altitude for
a subsonic flying vehicle anyway, so that refuelling hardly makes sense.
